Resumo: The oral squamous cell carcinoma (OSCC) the most frequent malignant cancer of head and neck region. It is considered a public health problem worldwide due to its high mortality. The AG is a phenolic compound having antiproliferative and cytotoxic properties presented in various tumor cells, but its role in OSCC is not fully understood. MET is a drug called insulin sensitizer responsible for activating protein kinase adenosine monophosphate (AMPK), which when activated, inactivates the mTOR pathway and activates the tumor suppressor protein involved in cell cycle control, p53 causing an antineoplastic effect important in tumor cells. The study examined the antineoplastic effect of gallic acid and of metformin on OSCC. They were evaluated proliferation, migration, invasion, apoptosis and the expression of certain genes involved in cancer. The AG and MET promoted decrease in the proliferation, migration and invasion and promote changes in genes such as E-cadherin, lactate and privutato dehydrogenase, caspase-3, HIF-1α in OSCC-9 compared with the immortalized keratinocytes (Hacat) . In conclusion, the results demonstrate that both AG and MET represent important as drugs that may be therapeutic strategies for the oral cancer
